TRIAL DESCRIPTION
Public title A Multicenter, Randomized, Double-blind, Placebo-controlled 12-week Study to Evaluate the Pharmacokinetics, Pharmacodynamics, and Safety of Oral Difelikefalin in Subjects with Advanced Chronic Kidney Disease and Moderate-to-Severe Pruritus with a 24-Week Open-Label Extension
Official scientific title A Multicenter, Randomized, Double-blind, Placebo-controlled 12-week Study to Evaluate the Pharmacokinetics, Pharmacodynamics, and Safety of Oral Difelikefalin in Subjects with Advanced Chronic Kidney Disease and Moderate-to-Severe Pruritus with a 24-Week Open-Label Extension
Brief summary describing the background and objectives of the trial This is a double-blind, randomized, placebo-controlled, multicenter study to evaluate the PK, PD, exposure-response relationship, and safety of oral difelikefalin in subjects with advanced CKD and moderate-to-severe pruritus. The study includes a Double-blind Treatment Period, an Open-label Extension Period, and a Follow-up Period. Double-blind Treatment Period Primary Objectives: To characterize the pharmacokinetic (PK) profile of oral difelikefalin in subjects with advanced chronic kidney disease (CKD) and moderate-to-severe pruritus. To determine exposure-response relationship of oral difelikefalin in subjects with advanced CKD and moderate-to-severe pruritus To evaluate the pharmacodynamic (PD) profile of oral difelikefalin in subjects with advanced CKD and moderate-to-severe pruritus based on change from baseline in weekly mean of the daily 24-hour WI-NRS score Secondary Objectives:To evaluate the PD profile of oral difelikefalin in improving sleep and global health measures in subjects with advanced CKD and moderate-to-severe pruritus To evaluate the safety and tolerability of oral difelikefalin in subjects with advanced CKD and moderate-to-severe pruritus Open-label Extension Period: Primary Objective:To evaluate the safety and tolerability of oral difelikefalin at a dose of 1 mg for up to 24 weeks of open-label treatment in advanced CKD subjects with moderate-to-severe pruritus
